Mechanisms for NR4A3 protein involvement during IT-induced cell death. B76 cells were seeded in 25 cm2 flasks and treated for 24 h with IT (10 ng/ml), CsA (2 μM) or the combination when cells were 70% confluent. Cell lysates were prepared and 15 μg total protein added each SDS-PAGE lane (A). The apparent molecular weight for the main band is 60 kDa and around 55 kDa for the lower band. In (B) cells were seeded in 75 cm2 flasks and treated as in (A). Adherent cells were used for subcellular fractionation. Immunoblots were probed with the NR4A3 antibody or subcellular fraction marker antibodies as indicated. The immunoblots are from one representative experiment of three independent experiments.
