Insulin and IGF-I increase OSE proliferation in a dose- and time-dependent manner. Ovarian organoids were cultured for 7d with increasing concentrations of insulin (A) or IGF-I (B) and analyzed for CK8 and BrdU expression. The percentage of OSE with BrdU incorporation was determined by dividing the number of cells positive for CK8 and BrdU by the total number of CK8-positive cells. C, Organoids were cultured for 1, 3, 7, or 14d in basal media (black line), media with 5 μg/ml insulin (blue line), or 5 μg/ml IGF-I (red line) and assessed for the percentage of OSE with BrdU incorporation as described above.
