Graphical representation of Ad vectors used in the study. The generated CRAd vectors have a deletion of 24 nucleotides (Δ24) in the early E1A gene (E1A) to allow selective replication in tumor cells with an pRb mutation. All three vectors encode a chimeric fiber protein (5/3 fiber) containing tail and shaft regions of Ad5 fiber fused with knob domain of Ad3, which is known to improve infection efficiency of OvCa cells. CRAd-IL24 (Ad5/3Δ24cmvIL24) contains IL-24 gene incorporated in place of the deleted E3B region (ΔE3B) under transcriptional control of the human CMV promoter (CMVp). CRAd-ING4 (Ad5/3Δ24cmvING4) is armed with CMVp-driven Inhibitor of Growth 4 (ING4) gene. Non-armed CRAd control was constructed to express reporter gene encoding the secreted Gaussia Luciferase (Gluc)
