Schematic of proposed mechanism for PDE10A inhibition anti-tumor effects in ovarian cancer. Elevated PDE10A alters the tumor microenvironment by influencing hormonal and inflammatory signaling. MCI-030 inhibits PDE10A in ovarian cancer cells resulting in increased cAMP and cGMP levels and activation of protein kinases, PKA and PKG, respectively. PKG inhibits β-catenin, decreasing nuclear translocation and transcription of downstream TCF/LEF targets. PKA may inhibit RAS signaling decreasing ERK and AKT activation
