The mechanism of the TP63-truncating mutation leading to POI. (A) The ATM-TP63 pathway regulates TP63 activity in healthy ovarian cells. However, when the TP63-truncating variant (TP63 trun) was generated, it activated the downstream target gene CLCA2, thereby stimulating its transcription and expression. This, in turn, induced more cell apoptosis, ultimately leading to POI. (B) The silencing of CLCA2 or the use of ATMi significantly inhibited the apoptotic effect of the TP63-truncated protein and decreased cell apoptosis
